By using this supplementation guide, you will be able to roll safley all while avoiding MDMA's well documented neurotoxicity. You will also learn how these suplements work to combat MDMA's negative effects on a fundemental level.

Why Use Supplements?

If the answer to this question is not obvious to you then thank God you're reading this guide. This concerns every MDMA user.

MDMA is an amazing and very safe substance if used properly. However, if used at high doses in sub-optimal environments, MDMA can cause neurotoxicity which literally means, it kills brain cells. In addition, supplementation can reduce the risk of losing the magic! Yup, you can almost entirely avoid loosing the magic by using supplements and smart dosing.

Listed below are the supplements one should take and how to take them.

Alpha Lipoic Acid (ALA)

Alpha Lipoic Acid or ALA for short is a metabolic anti-oxidant that is a must-have in your supplementation regimen as ALA was found to work wonders at preventing MDMA's neurotoxicity.

This can be seen in the quote from a study below.

“Repeated administration of the metabolic antioxidant alpha-lipoic acid (100 mg/kg, i.p., b.i.d. for 2 consecutive days) 30 min prior to MDMA did not prevent the acute hyperthermia induced by the drug; however, it fully prevented the serotonergic deficits and the changes in the glial response induced by MDMA.”

Optimal dosage schedule:

  • 300mg Alpha Lipoic Acid with MDMA
  • Re-dose 300mg every 2 hours until 7 hour mark
  • Do not consume any more than 2400mg per day

Acetyl-L-Carnitine (ALCAR)

ALCAR is naturally produced in the body although it is most often taken as a dietary supplement to maximise its benefits.


ALCAR works by being broken down in the blood into carnitine which is used by the body as a means of transporting fatty acids into mitochondria for energy.

A study found that ALCAR provided in vivo neuroprotection against MDMA's mitochondrial neurotoxicity. Here is a summary of the findings.

The present work is the first to successfully demonstrate that pretreatment with ALC exerts effective neuroprotection against the MDMA-induced neurotoxicity at the mitochondrial level, reducing carbonyl formation, decreasing mtDNA deletion, improving the expression of the respiratory chain components and preventing the decrease of 5-HT levels in several regions of the rat brain.”

Optimal dosing schedule:

  • 500mg 3 hours before MDMA
  • 500mg 1 hour after MDMA
  • 500mg 5 hours after MDMA
  • Do not consume more than 2500mg of ALCAR per day

Electrolytes and Isotonic Fluids

MDMA is well-known ability to dehydrate the user and make them unable to urinate. Additionally, MDMA causes hyponatremia which is a low level of sodium in the user's blood. It can lead to symptoms like decreased ability to think, headaches and nausea all the way to seizures and death. Due to this, **staying hydrated while rolling is crucial. **


Regular water is usually not good enough while rolling, this is due to MDMA potentiating water's ability to lower sodium in the blood serum.

Prevention of hyponatremia with limited consumption of electrolyte containing fluids and controlled ambient temperatures are required to preserve the body’s homeostatic maintenance of fluid balance.”

Optimal dosing schedule:

  • ~110mg of sodium with 250ml of fluid an hour if not dancing
  • 500ml of fluid an hour if dancing
  • Do not consume more than 2300mg of sodium per day

Vitamin C

Vitamin C, also known as ascorbic acid, is a vitamin found in food and is widely used as a dietary supplement. Supplementation of vitamin C is used to treat and prevent scurvy. It is also an anti-oxidant hence why it can help with MDMA neurotoxicity.


Below is a quote from a study that shows benefits of using Vitamin C with MDMA.

“Treatment of rats with ascorbic acid suppressed the generation of hydroxyl radicals, as evidenced by the production of 2,3-dihydroxybenzoic acid from salicylic acid, in the striatum during the administration of a neurotoxic regimen of MDMA. Ascorbic acid also attenuated the MDMA-induced depletion of striatal 5-HT content.”

This means that vitamin C suppressed the creation of radicals which are molecules that cause oxidative stress to the CNS, thus lowering neurotoxicity.

Optimal dosing schedule:

  • 500mg every 3 hours
  • First dose ~3 hours before taking MDMA
  • Last dose ~6 hours after MDMA.
  • Use caution as doses above 2,000-6,000mg may cause diarrhoea.

Magnesium (Optional)

Magnesium is thought to be a weak NMDA antagonist and is thought to reduce jaw clenching in anecdotal reports. It does not offer neuroprotection like the other supplements.

Optimal dosing schedule:

  • 1-2 tablets (100-200mg magnesium) taken as needed

5-HTP (Optional)

Anecdotal reports show that 5-HTP supplementation may be effective at reducing the severity of MDMA comedowns as it acts as a precursor to serotonin which gets's depleted as a result of MDMA administration

Optimal dosing schedule:

  • 100mg up to 3 times a day
  • While 5-HTP is usually well tolerated, high doses can cause nausea and unwanted peripheral side effects.
