The first step relies on the fact that most eukaryotic mRNAs have poly(A) tails at their 3′ ends. The poly(A) tails serve as "hooks" during the separation process. This process involves pouring all of the cellular RNA (tRNA, rRNA, snRNA, mRNA, etc.) into what is known as an elution column of short DNA pieces, mostly thymine nucleotides.
